Esomeprazole 40mg is indicated for the targeted treatment of severe acid-mediated gastrointestinal conditions:
Healing of Erosive Esophagitis: 4-to-8-week short-term therapy to heal acid-induced esophageal lesions.
Maintenance of Healed Esophagitis: Continuous therapy to prevent the recurrence of esophageal erosions.
Symptomatic GERD: Relief of persistent, severe heartburn when lower doses fail.
H. pylori Eradication: Used in combination with antibiotics to eliminate H. pylori and prevent duodenal ulcers.
Hypersecretory Conditions: Long-term management of severe acid overproduction, such as Zollinger-Ellison Syndrome.
Dosage & Administration
Website Note: Dosage must be individualized by a registered medical practitioner based on the patient’s specific diagnosis.
Standard Dose: Typically 40mg taken orally once daily for 4 to 8 weeks (or twice daily for Zollinger-Ellison Syndrome).
Administration: Tablets must be swallowed whole with water—do not crush or chew.
Timing: Must be taken at least one hour before a meal (preferably before breakfast).
Contraindications & Precautions
Absolute Contraindications: Do not use in patients with known hypersensitivity to esomeprazole or substituted benzimidazoles. Co-administration with rilpivirine is strictly contraindicated.
Long-Term Risks: Prolonged use (over 1 year) may increase the risk of osteoporosis-related bone fractures and severe low magnesium levels (hypomagnesemia).
Infection Warning: Reduced gastric acid may increase the risk of Clostridium difficile-associated diarrhea (CDAD).
Masking Symptoms: Symptomatic relief does not rule out underlying gastric malignancy; severe symptoms (e.g., unexplained weight loss) require medical screening.
Potential Side Effects
Common / Mild: Headache, diarrhea, abdominal pain, nausea, flatulence, and constipation.
Less Common / Severe: Chronic Vitamin B-12 deficiency (with greater than 3 years of daily use), acute kidney reaction (interstitial nephritis), and muscle spasms/arrhythmias due to low magnesium.
